Sperm DNA Fragmentation: What It Means for Fertility

Sperm DNA fragmentation is the proportion of breaks in the genetic material that sperm carry. A sample can look normal on a standard semen analysis (count, motility, shape) and still have a high level of DNA fragmentation.

What is sperm DNA fragmentation?
During fertilisation, sperm carry the father's genetic information into the egg. When breaks form in the DNA strands that hold this information, it is called DNA fragmentation.
A low level exists in most men; higher levels can affect fertilisation and embryo development.
Causes and risk factors
Factors that can raise DNA damage include:
- Smoking and heavy alcohol use
- Advanced age and prolonged febrile illness
- Varicocele
- Environmental toxins and excessive heat exposure
- Oxidative stress and an unbalanced diet
How is it measured?
DNA fragmentation is assessed on a semen sample with specialised laboratory tests and reported as a percentage (index). It does not replace the standard semen analysis; it complements it.
